A Study on Strain Improvement by Protoplast Fusion between Amylase Secreting Yeast and Alcohol Fermenting Yeast


Abstract
In the alcohol industry, the substrate of starch is not directly fermentable material by yeast and must be pretreated to hydrolyze it to simple sugars by diastatic enzymes obtained from molds or plant. Futhermore, the capital investment for the pretreatment of starch material was actually one of the main causes to increase the alcohol plant cost, much efforts were devoted to develope a new brewing yeast capable of fermenting starch directly to alcohol. In our recently work, we attempted intergeneric protoplast fusion between amylase secreting yeast(S. diastaticus) and alcohol fermenting yeast (S. cerevisiae) to breed a new brewing yeast. The successful fusion products were capable of using starch and producing alcohol from preliquefied potato starch(D.E=3.0) with high efficiency. The characteristics and genetic stability of fusants were also investigated.
In this study we have focused on the investigation of the optimum conditions of alcohol fermentation in laboratory scale and in pilot plant scale to estimate the possibility of industrial application of the fusants.
